A Magical Girl (魔法少女?) or otherwise known as Puella Magi in the mangas and other official media is a girl who has contracted with a messenger of magic known as Kyubey, having one wish granted and in exchange, the contractees must fight, battle and exorcise Witches. Later, it is revealed that Magical Girls are actually meant to rebirth into and become Witches someday, when all their hopes and dreams will be lost and destroyed.

- According to Kyubey in episode 5, a Magical Girl will receive one unique ability based on the theme of their wish.
- Starting with the same episode, a Magical Girl's motif will appear as a mark on the nail of her middle finger.
- According to Kyubey in episode 6, if a Magical Girl is 100 meters away from her Soul Gem, she will become unconscious until she touches her Soul Gem. Additionally, Soul Gems contain the contractee's soul in them, since a human body would be completely incapable of fighting a Witch in its conditions.
- According to Kyubey in episode 8, a teenager who is on their way to become a woman is simply named "girl". Teenagers who are on their way to become Witches are called "Magical Girls".
- According to Kyubey in episode 11, the potential of a girl becoming a Magical Girl is evolved from her life conditions or circumstances that surround her. Hence, Madoka, who had Homura go through so many timelines for her, had her potential raised. In the same episode, the reason why Magical Girls are born is given. Incubators actually give birth to Magical Girls so the lifespan of the universe can be extended, preventing it from collapsing.
- Madoka and Homura are the only known Magical Girls who are confirmed to have made two or more different wishes in all timelines.
